of APP expression and processing in vivo, we used brain and cerebral microvessels from eNOS-deficient (eNOS / ) mice. Brain tissue from eNOS / mice had statistically higher APP and BACE1 protein levels, as well as increased BACE1 enzyme activity and A (A 1-42 wild-type control, 0.737pg/mg; eNOS / , 1.475 pg/mg; P<0.05), compared with wild-type controls (n 6 to 8 animals per background). Brain microvessels from eNOS / mice also showed statistically higher BACE1 protein levels as compared with wild-type control. Conclusions: Our data suggest that endothelial NO plays an important role in modulating APP expression and

Endothelial nitric oxide modulates expression and processing of amyloid precursor protein.

RATIONALE the exact etiology of sporadic Alzheimer disease (AD) is unclear, but it is interesting that several cardiovascular risk factors are associated with higher incidence of AD. The link between these risk factors and AD has yet to be identified; however, a common feature is endothelial dysfunction, specifically, decreased bioavailability of nitric oxide (NO). OBJECTIVE to determine the ...

Investigation of the Iron Oxide Nanoparticle Effects on Amyloid Precursor Protein Processing in Hippocampal Cells

Introduction: Iron oxide nanoparticles (Fe2O3-NPs) are small magnetic particles that widely used in different aspects of biology and medicine in modern life. Fe2O3-NP accumulated in the living cells due to absence of active system to excrete the iron ions so damages cellular organelles by highly reactivity. Method: Herein cytotoxic effects of Fe2O3-NP with 50 nm size were investigated on prima...

Oxidative stress affects processing of amyloid precursor protein in vascular endothelial cells

BACKGROUND Oxidative stress is thought to be a key player in the pathogenesis of neurodegenerative dementia, including Alzheimer's disease (AD). It has been assumed that oxidative stress contributes to the ß-amyloid deposition in cerebral blood vessels. METHODS In order to prove this hypothesis, we examined the effect of oxidative stress on the processing of amyloid precursor protein (APP) in...

Absence of Nitric Oxide Synthase 3 Increases Amyloid β-Protein Pathology in Tg-5xFAD Mice.

AIM The abnormal accumulation, assembly and deposition of the amyloid β-protein (Aβ) are prominent pathological features of patients with Alzheimer's disease (AD) and related disorders. A number of factors in the brain can influence Aβ accumulation and associated pathologies. The aim of the present study was to determine the consequences of deleting nitric oxide synthase (NOS) 3, the endothelia...

Effects of Nerve Growth Factor and Nitric Oxide Synthase Inhibitors on Amyloid Precursor Protein mRNA Levels and Protein Stability

We determined previously that nitric oxide (NO) modulates the nerve growth factor (NGF)-mediated increases in amyloid precursor protein (APP) levels in PC12 cells. To elucidate potential mechanisms, the effects of NGF and NO synthase (NOS) inhibitors on APP mRNA levels and protein stability were evaluated. Surprisingly, treatment of PC12 cells with NGF resulted in decreased levels of APP695 and...
